 CULTURE MEDIUM AND CULTURE MEDIA 

CULTURE MEDIUM:

DEFINITION: A chemically prepared solid /liquid substance used to support the growth of microorganism

Uses : 

  1. To provide nutrition
  2. To provide space
  3. To provide growth
  4. To increase population
  5. To handle stress
  6. To differentiate species 


COMPONENTS:

1) NITROGEN SOURCE- Amino,protein extract,peptone

2) Growth factor- Blood serum,meat,yeast extract

3)Energy- Sugar,alcohol,carbohydrate

4)Buffer - Maintain ph ,phosphate,acetate 

5) Salt- Enhance growth,phosphate ,sulfate,magnesium



SELECTIVE AGENTS:

  • CHEMICAL
  • ANTIBIOTICS
  • INDICATOR DYE

GELLING AGENT : 

  • Act as solidifying agent 
  • agar ,gelatin- agents
CULTURE MEDIA 

  • Culture media is required to grow organism from infected material to identify the causative agent
  • It contains: water,vitamin,mineral,growth factor,blood,serum,agar,peptone  
TYPES :
  1. SIMPLE
  2. COMPLEX
  3. SPECIAL: ENRICHED,ENRICHMENT,SELECTIVE,DIFFERENTIAL,INDICATOR,TRANSPORT,SUGAR 
  4. SYNTHETIC
  5. ANAEROBIC : DEHYDRATE 


SIMPLE:
  • Nutrient broth - peptone water+meat extract
  • glucose broth- peptone water+glucose
  • nutrient agar- nutrient broth +agar ( 2-3%)
complex media : 
  • all media other than simple media
synthetic media: 
  • they are prepared from pure chemical
media                                          use 
  • nutrient broth                  routine culture
  • glucose broth                  blood culture
  • nutrient agar                routine culture 
SPECIAL MEDIA: 
BLOOD MEDIA - Nutrient agar +sheep blood
eg- streptococcus -require blood for growth
CHOCOLATE AGAR;
Used in haemophilus influenza
ENRICHMENT MEDIA 
1) SELECTIVE F BROTH
Culture of faeces for salmonella and shigella
2) cooked meat broth /robertson cooked meat broth
culture of anaerobic bacteria
SELECTIVE MEDIA : 
  1. Deoxycholate citrate - culture of salmonella ,shigella
  2. bile salt agar - culture of vibrio cholerae





INDICATOR MEDIA 
Macconkey's medium- culture of gram negative bacilli
TRANSPORT MEDIA:
  • They are used for the transport of the clinical sample suspected to contain delicate organism or when the delay is expected while transporting the sample
  • Simpler terms from site of collection to laboratory 
  • Condition in the media is in such a way that they don't multiply within the collected container 
  • This also prevents drying of sample and maintain viability of the organism in the sample
  • Examples: stuart medium for neisseria species,
  • venkatraman ramakrishnan medium -vibrio cholerae
